My stress related to my mental illnesses is higher than ever. I can hardly think anymore so doing research on methods like how to tie a rope just right is making my brain go Blue screen.

There was a spate of suicides in Turkey sometime back because cyanide was freely available. The Turkish authorities restricted public access to cyanide and the suicide rate dramatically dropped. It really does take a lot of courage to kill yourself with the available methods which is why the suicidal are often misperceived as violent and unbridled.

And how do you tie the other end without it coming undone? Where in a hotel room can I do this at? I was thinking of using duct tape to reinforce wherever I tie the rope.

What kind of rope? I know not one that's stretchy.

Use a snuggle hitch, as described here - https://en.wikipedia.org/wiki/Snuggle_hitch
Not sure how you would exactly go about doing it in a hotel, but I've heard of people using doors and windows. For the rope, I have a 12mm and 10mm double braided polyester rope. If I hang, I'll be using a tree or other sturdy anchor point. Maybe someone else can weigh in, no pun intended.

EDIT: I meant polyester, not nylon

not sure, the casino near me has very strong bars, could hold me up. id never do it though, those maids do enough.

If you go to a nicer hotel things are likely to be stronger. If you're doing full you might be better finding a tree than trusting a hotel. If you're doing partial most nice hotels should have plenty of places that can handle your weight. Always strength test with your arm before your neck.

How? a rope seems better except for the part where the other end of the rope needs to be tied well to the anchor.

I was at Home Depot today and the rope available that wasn't nylon or abrasive was 100ft bundles. What length would I cut it to?
Even a moderately nice place. Just not a road side shack. As for rope length I don't know. It depends on your hight and anchor points hight. Just bring something to cut it.

What would be the best knot for a long drop of 15 feet +to pretty much guarentee neck snaps. Every hangmanscnoosecis different methods people tweak themselves. You feel like Tom hanks in cast away testing the noose.

As long as the rope slides and allows you to adjust the size of the noose without becoming undone, then it's correct. Try to make the noose as small as you can. If the whole thing comes undone, then it's not correct.
Why do you use two different ropes? Or did you just get two so you can choose which one is better?
Just so I could compare. By most accounts, you don't want to go too thick as it won't constrict as well. I'd say 10mm maximum. Really thick ropes are used in drop hangings to break the neck.
