I plan to take one of the guns from my uncle's house and shoot myself. I'd feel bad dying over at his house, like yeah, what a way to show thanks for someone's hospitality. I don't want them to have to hear the gunshot and discover my mangled body. They're stressed out enough. I just want this to be as stress-free as possible for everyone involved.

I don't think shooting a gun in a hotel is the safest idea, but no, they wouldn't check your luggage.

I guess it may depend on where you are maybe? I've traveled internationally and never had a hotel ask to check my luggage but I hadn't been to communist or middle eastern countries so I don't have 1st hand experience with their rules or laws. Sorry.

Make sure the rules of your area don't hold the owners of the gun accountable for your suicide, sound and aftermath of your body could be the least of their problems.

You can always say no. No hotel security can forcibly search you or your belongings. They may not let you stay there if you refuse, but you can just move on to another hotel. The hotel's policies should be clearly indicated on their website or premises signage.

I have never heard of a hotel searching anyone's bags. That would be a MASSIVE privacy violation and probably illegal. If they were suspicious, they would probably call the police to search your bags, but as long as you act like a normal person and don't threaten anyone or walk in with obviously gun-shaped bags, I don't think anyone will notice or care.

A hotel would have no legal right to search bags unless they go through the hassle of calling cops or denying you service.

Otherwise, as long as you more-or-less conceal it you should be fine. Alot harder with a long-gun, but I think using a golfing or duffel bag for carrying sports equipment would also work, esp if it's apparent if it's what it's for. I really don't think "Gun-shaped bag" is too conspicuous given an adequate cover for just staying at a motel. What would raise eyebrows is bringing actual rifle/gun storage like large cases or bags made specifically to carry guns.

If it's a hand-gun, you could just put it in any bag.

Ofc, it'd be just a matter of not looking soo nervous, shady, suspicious, etc. But relatively easy, or should be.

Also, since you're going for gun, I'd like to ask why'd you'd be doing it in a hotel and not outside? The *bang*, and mess could be a hassle for the whole building. I think gun CTB somewhere in the woods would be better but that's just me.
